Epidemiologic research into trehalase enzymopathy experienced a significant advancement when the correlation between reduced trehalase activity and the A allele of the tTREH gene (rs2276064) became apparent. This study investigated the allelic and genotypic frequencies of the trehalase gene in indigenous populations of Siberia and the Russian Far East. A reference dataset consisting of 567 samples representing indigenous populations of Siberia and the Russian Far East, along with 146 samples from Eastern Slavs, was used for genotyping. An increase in A*TREH allele frequencies was observed in an eastward direction, as our investigation demonstrated. In the reference group, the A*TREH allele's frequency was 0.003. North-West Siberian indigenous groups exhibited a frequency between 0.013 and 0.026. The A*TREH allele frequency ranged from 0.029 to 0.030 in South Siberia. West Siberia exhibited a frequency of 0.043, while the low Amur populations showed a frequency of 0.046. The A allele (063) exhibited its highest frequency in the populations of Chukchi and Koryak. Amongst those of European heritage, the risk of trehalase enzymopathy is observed in a population segment ranging from 1% to 5%. Novel biomarkers for vascular disease in diabetes may illuminate previously unknown mechanistic pathways. The multifaceted process of bone and vascular calcification, involving osteocalcin, osteoprotegerin, and osteopontin, is often compromised in those with diabetes. Possible connections between osteocalcin, osteoprotegerin, and osteopontin and cardiovascular disease (CVD) and diabetic retinopathy (DR) were examined in a population of type 2 diabetes (T2D) patients.
The SUMMER Study, involving 848 participants with type 2 diabetes, measured the levels of osteocalcin, osteoprotegerin, and osteopontin at the commencement of the trial, as documented on ClinicalTrials.gov. Employing logistic regression models in conjunction with propensity score matching, we investigated potential associations between a history of CVD and evidence of any grade of DR, and osteocalcin, osteoprotegerin, and osteopontin, while adjusting for influencing factors.
Participants with a prior history of CVD numbered 139 (164%), in contrast to 144 (170%) who exhibited DR. After accounting for potential confounding variables, a correlation was found between osteocalcin concentrations and a history of CVD, but not between osteoprotegerin or osteopontin concentrations and CVD. The odds ratio (OR) and 95% confidence interval (CI) for a one standard deviation (SD) increase in the natural log of osteocalcin concentrations was 1.35 (1.06-1.72), p=0.0014. Benzylamiloride Osteoprotegerin and osteopontin levels were found to be associated with prevalent DR, while osteocalcin was not. A one standard deviation rise in osteoprotegerin (natural log) was associated with a 1.25-fold increase in odds of prevalent DR (95% CI: 1.01-1.55, p=0.0047). Likewise, a one standard deviation rise in osteopontin (natural log) corresponded to a 1.25-fold increase in odds (95% CI: 1.02-1.53, p=0.0022).
Macrovascular complications in T2D are linked to elevated serum osteocalcin levels, while microvascular complications correlate with higher osteoprotegerin and osteopontin concentrations, implying a potential role for these osteokines in vascular disease pathways.
